目的观察研究二乙氧基乙醇(2-Ethoxyethanol,2-EE)染毒雄性大鼠睾丸和附睾毒性、精子毒性、以及性行为与妊娠率的改变,探讨各生殖毒性指标之间的关联性,为确定2-EE雄性生殖毒性健康监护指标和诊断指标,开展2-EE职业健康危险度评价提供科学依据。方法选择清洁级雄性Wistar大鼠,分别给予剂量200、400和800 mg/kg的2-EE,连续灌胃13 d,观察其睾丸和附睾脏器系数、病理改变;精子计数和精子活力改变;染毒雄鼠与正常雌鼠合笼,观察其性行为和配对雌鼠妊娠率的改变。对照组以等量生理盐水灌胃。结果 800 mg/kg染毒组睾丸脏器系数明显下降(P<0.01),各级生精细胞明显减少,精子活力下降(P<0.05);400 mg/kg以上染毒组雄鼠骑跨潜伏期、插入潜伏期、射精潜伏期和射精后间隔期不同程度延长,差异无显著性统计学意义(P>0.05);雌鼠妊娠率与对照组比较差异无显著性统计学意义(P>0.05)。结论 2-EE染毒雄鼠睾丸毒性明显,精子毒性表现滞后于性行为改变且毒效应剂量较高,性行为改变可作为2-EE雄性生殖毒性直观而敏感的效应指标。
Objective To investigate the changes of testis and epididymitis, sperm toxicity, sexual behavior and pregnancy rate in male rats exposed to 2-Ethoxyethanol (2-EE), and to explore the correlation between reproductive toxicity indexes, To determine 2-EE male reproductive toxicity of health monitoring indicators and diagnostic indicators, to carry out 2-EE occupational health risk assessment provide a scientific basis. Methods Clean male Wistar rats were randomly divided into two groups: 200, 400 and 800 mg / kg 2-EE, respectively. The rats were given gavage for 13 days. The organ coefficients of testis and epididymis, pathological changes, sperm counts and sperm motility were observed. The male and female mice were caged to observe the change of their sexual behavior and the pregnancy rate of the matched female rats. Control group with the same amount of saline gavage. Results The organ coefficient of testis significantly decreased (P <0.01) at 800 mg / kg exposure, the spermatogenic cells at all levels significantly decreased, and the sperm motility decreased (P <0.05) (P> 0.05). There was no significant difference in the pregnancy rate between the female mice and the control group (P> 0.05). Conclusions The testicular toxicity of 2-EE in male rats is obvious. The sperm toxicity lags behind the change of sexual behavior and the dose of toxic effect is high. The change of sexual behavior can be used as an intuitive and sensitive effect indicator of 2-EE male reproductive toxicity.
