为探讨烧伤早期内皮素变化及不同浓度ET-1对培养血管内皮细胞通透性的影响,动态观察了大鼠30%Ⅲ度烧伤早期血浆内皮素变化规律及其对内皮细胞通透性的直接影响。结果表明:血浆ET浓度伤后迅速升高,6小时达峰值,以后虽有所回落,但24小时内仍显著高于基础水平(P<0.01);培养血管内皮细胞单层与0,1,10,100nmol/L ET-1孵育后,其对Hank’s液或白蛋白灌注液的滤过流量、滤过系数均明显增加,而对蛋白渗透压反射系数显著减少,且存在剂量依赖性。提示烧伤后异常增高的ET除发挥其强大的缩血管作用,调节血流分布外,还直接促进了烧伤后毛细血管通透性的增加。
In order to investigate the changes of endothelin in early stage of burn and the effect of different concentrations of ET-1 on the permeability of cultured vascular endothelial cells, the changes of plasma endothelin in early stage of 30% burn and the permeability to endothelial cells influences. The results showed that the concentration of plasma ET increased rapidly after injury and peaked at 6 hours, although it dropped after 24 hours, it was still significantly higher than the baseline within 24 hours (P <0.01) After incubating with 10,100nmol / L ET-1, the filtration rate and filtration coefficient of Hank’s solution or albumin perfusion solution increased significantly, but the osmotic pressure reflection coefficient decreased significantly in a dose-dependent manner. Tip abnormally elevated ET in addition to exert its powerful role in vasoconstriction, regulating blood flow distribution, but also directly contribute to the increase of capillary permeability after burn.
