目的:采用戊型肝炎病毒主要免疫表位多肽检测特异性IgM抗体。方法:采用戊型肝炎病毒主要免疫表位多肽,制备ELISA检测试剂,同时使用北京万泰生物药业公司生产的抗-HEV IgM抗体诊断试剂,分别检测33例戊型肝炎患者血清和344例普通人群血清抗-HEV IgM,并对部分阳性标本用免疫印迹法(Western blot,WB)确认。结果:用本室ELISA试剂检测33例戊型肝炎患者血清,抗-HEV IgM均阳性,符合率为100%;用万泰公司试剂检测阳性31例,符合率为93.9%。344例普通人血清中,本室试剂检测抗-HEV IgM阳性11例,而用万泰试剂检测仅其中6例显示阳性;对本室试剂阳性而万泰试剂阴性的5例进一步用WB检测,抗-HEV IgM均阳性。结论:本室制备的ELISA试剂可准确高效地检测血清抗-HEV IgM。
OBJECTIVE: To detect specific IgM antibodies using hepatitis E virus epitopes. Methods: The main epitope peptide of hepatitis E virus was used to prepare the ELISA test reagent. At the same time, the anti-HEV IgM antibody diagnostic reagent produced by Beijing WANTAI Biopharmaceutical Co. was used to detect the serum of 33 hepatitis E patients and 344 normal Serum anti-HEV IgM was detected in the serum and some positive samples were confirmed by Western blot (WB). Results: The serum and anti-HEV IgM of 33 patients with hepatitis E were detected by our ELISA kit, and the coincidence rate was 100%. The positive rate was 93.9% in 31 cases detected by Wantei reagent. Of 344 normal human serums, 11 were positive for anti-HEV IgM in our laboratory, whereas only 6 of them were positive in our test; 5 were negative for this agent and 5 were further tested by WB for anti-HEV IgM -HEV IgM are positive. Conclusion: The ELISA reagent prepared in our laboratory can detect serum anti-HEV IgM accurately and efficiently.
