神经细胞骨架作为重要的结构和功能蛋白,在神经损伤、疾病的研究中具有重要意义。本研究利用α管蛋白的单抗及其互补DNA做探针,借助于免疫组化、原位杂交、图像分析等手段,对大鼠脊髓缺血损伤后不同时点损伤脊髓α管蛋白及其mRNA进行定性定量研究。结果显示,α管蛋白及其mRNA的变化相同步,伤后1天两者都明显降低,3天达最低水平,7天有所恢复,14天基本恢复到伤前。脊髓损伤后α管蛋白表达的变化,在时间上与继发性脊髓损伤相一致,表明各种继发性损伤机制可能参与细胞骨架的变化,如Ca2+激活的中性蛋白酶等
As an important structural and functional protein, neural cytoskeleton plays an important role in the study of nerve injury and disease. In this study, the α-tubulin monoclonal antibody and its complementary DNA were used as probes. The expression of α-tubulin in spinal cord at different time points after spinal cord ischemia injury was detected by immunohistochemistry, in situ hybridization and image analysis mRNA qualitative and quantitative research. The results showed that the changes of α-tubulin and its mRNA were synchronized. Both of them were significantly reduced on the 1st day after injury, reaching the lowest level on the 3rd day, recovering on the 7th day and recovering to the baseline on the 14th day. Changes in the expression of α-tubulin after spinal cord injury coincide with secondary spinal cord injury in time, indicating that various secondary mechanisms of injury may be involved in changes in the cytoskeleton, such as Ca2 + -activated neutral proteases
