目的研究吗啡条件性位置偏爱期间疏肝解郁方对大鼠前额叶皮质和伏核中谷氨酸受体1(GluR1)、谷氨酸受体2(GluR2)蛋白表达的影响。方法采用Western blot实验方法定量检测GluR1和GluR2蛋白表达的变化。结果与正常对照组比较,吗啡模型组GluR1和GluR2的蛋白表达均明显升高(P<0.01)。与吗啡模型组比较,疏肝解郁方对吗啡模型组GluR1和GluR2的蛋白表达均明显下调,差异具有统计学意义。结论吗啡模型组大鼠前额叶皮质和伏核GluR1和GluR2的蛋白表达增多,参与了吗啡依赖的形成过程。而疏肝解郁方对前额叶皮质和伏核GluR1和GluR2的蛋白表达具有明显下调作用,干预了吗啡依赖的形成。
Objective To study the effect of Shugan Jieyu Decoction on glutamate receptor 1 (GluR1) and glutamate receptor 2 (GluR2) protein expression in prefrontal cortex and nucleus accumbens during morphine conditioned place preference. Methods Western blot was used to detect the expression of GluR1 and GluR2 protein. Results Compared with the normal control group, the protein expressions of GluR1 and GluR2 in morphine model group were significantly increased (P <0.01). Compared with morphine model group, the expression of GluR1 and GluR2 protein of Shugan Jieyu Decoction group were significantly decreased, the difference was statistically significant. Conclusion The expression of GluR1 and GluR2 in the prefrontal cortex and nucleus accumbens increased in morphine model rats, which is involved in the morphogenesis of morphine dependence. However, Shugan Jieyu Decoction significantly reduced the protein expression of GluR1 and GluR2 in prefrontal cortex and nucleus accumbens, which interfered with the morphine-dependent formation.
