目的:应用蛋白质组学的方法检测非酒精性脂肪肝大鼠肝脏线粒体中差异蛋白,探讨香砂六君丸治疗非酒精性脂肪肝的作用机制。方法:将24只雄性SD大鼠随机分为正常对照组、模型组与治疗组,每组8只。采用高脂饲料喂养法复制非酒精性脂肪肝模型,香砂六君丸(1.35 g/kg·d)灌胃4周治疗后取材,采用i TRAQ技术进行差异蛋白的检测。结果:与正常组大鼠相比,模型组大鼠线粒体蛋白表达具有统计学意义的差异蛋白共61个。与模型组大鼠相比,香砂六君丸组大鼠线粒体蛋白表达有统计学意义的差异蛋白共48个。3组大鼠共同的线粒体蛋白表达有统计学意义的差异蛋白有19个。结论:香砂六君丸可能通过调节线粒体能量代谢的机制以发挥治疗非酒精性脂肪肝的作用。
OBJECTIVE: To detect the differential proteins in liver mitochondria of non-alcoholic fatty liver rats by proteomics and to explore the mechanism of Xiangsha Liujun pill in treating non-alcoholic fatty liver. Methods: 24 male SD rats were randomly divided into normal control group, model group and treatment group, with 8 rats in each group. The model of non-alcoholic fatty liver was duplicated by high-fat diet feeding. Xiangsha Liujun pill (1.35 g / kg · d) was given for 4 weeks after gavage. The differential protein was detected by i TRAQ technique. Results: Compared with the normal group, the expression of mitochondrial protein in the model group was statistically significant differences in a total of 61 proteins. Compared with the model group, the expression of mitochondrial protein in Xiangsha Liujun pill group had 48 statistically significant differences. There were 19 differentially expressed proteins with common mitochondrial protein expression in the 3 groups. Conclusion: Xiangsha Liujun pill may play a role in the treatment of non-alcoholic fatty liver by regulating the mechanism of mitochondrial energy metabolism.
