银杏叶提取物新配方对高脂血症大鼠的干预效应(英文)

背景:调脂治疗在冠状动脉粥样硬化性心脏病的一、二级预防中的作用已得到充分肯定,银杏叶提取物具有一定的降脂作用。目的:建立高脂大鼠模型,观察银杏叶提取物新配方(由银杏总黄酮和银杏内酯组成)对高脂血症大鼠血脂水平的干预及肝脏脂肪变性程度。设计:随机对照观察。单位:复旦大学附属金山医院心内科,苏州大学药学系,苏州中药研究所。材料:选取清洁级 SD 大鼠 60只,适应性饲养 1周后,随机分成 6组:正常对照组、高脂模型组、力平脂 25m g/(kg·d)组、银杏叶提取物新配方20m g/(kg·d)组、银杏叶提取物新配方 40m g/(kg·d)组、银杏叶提取物新配方 80m g/(kg·d)组,10只 /组。方法:力平脂 25m g/(kg·d)组、银杏叶提取物新配方 20m g/(kg·d)组、银杏叶提取物新配方 40m g/(kg·d)组、银杏叶提取物新配方 80m g/(kg·d)组均以 10m L /kg 体积于每天上午 9:00~10:00灌服给药,正常对照组和高脂模型组给予等体积的蒸馏水灌服。除正常对照组外,其余各组大鼠于每天下午 1:00~2:00同时以 10m L /kg 体积灌服脂肪乳剂,连续 28d。第 29天各组分别测定血清总胆固醇、三酰甘油和高密度脂蛋白胆固醇、血清载脂蛋白 AI、血清载脂蛋白 B。同时取出各组大鼠肝脏进行病理检查,观察肝脏脂肪变性程度。主要观察指标:造模后各组大鼠的血脂情况以及肝脏脂肪变性程度。结果:实验选用大鼠 60只,均进入结果分析。①造模后各组大鼠血脂情况的比较:高脂模型组血清总胆固醇、低密度脂蛋白胆固醇明显增加,高密度脂蛋白胆固醇降低,与正常对照组比较差异显著(P <0.01),但血清三酰甘油仅有轻度升高(P >0.05);银杏叶提取物新配方 20~80m g/(kg·d)组均可不同程度地抑制大鼠高脂血症的形成,表现为血清总胆固醇、三酰甘油、低密度脂蛋白胆固醇均明显降低(P <0.05或 0.01),并有一定升高高密度脂蛋白胆固醇、血清载脂蛋白 AI及降低血清载脂蛋白 B 的作用,其中银杏叶提取物新配方 80m g/(kg·d)组最为显著(P <0.05)。②造模后各组大鼠肝脏脂肪变性程度的比较:高脂模型组大鼠的肝病理检查可见弥漫性脂肪空泡;银杏叶提取物新配方 40,80m g/(kg·d)组大鼠肝脂肪变性程度有一定的减轻,尤以银杏叶提取物新配方 80m g/(kg·d)组效果明显。结论:提示银杏叶提取物新配方可有效地纠正高脂血症的脂质代谢,对高脂饮食所致的高脂血症具有良好的预防作用。 BACKGROUND: The role of lipid-regulating therapy in the primary and secondary prevention of coronary atherosclerotic heart disease has been fully affirmed. Ginkgo biloba extract has a certain lipid-lowering effect. OBJECTIVE: To establish a high-fat rat model and observe the effects of a new formulation of Ginkgo biloba extract (composed of total flavones of ginkgo biloba and ginkgo lactone) on blood lipid levels in hyperlipidemic rats and liver steatosis. Design: Randomized controlled observations. Unit: Department of Cardiology, Jinshan Hospital, Fudan University, Department of Pharmacy, Soochow University, Suzhou Institute of Chinese Materia Medica. MATERIALS: A total of 60 clean grade SD rats were selected. After adaptive feeding for 1 week, they were randomly divided into 6 groups: normal control group, high-fat model group, Liping 25m g/(kg·d) group, and Ginkgo biloba extract. Formula 20m g/(kg·d) group, Ginkgo biloba extract new formula 40m g/(kg·d) group, Ginkgo biloba extract new formula 80m g/(kg·d) group, 10/group. Methods: Lipopenin 25mg/(kg·d) group, Ginkgo biloba extract new formula 20mg/(kg·d) group, Ginkgo biloba extract new formula 40mg/(kg·d) group, Ginkgo biloba extract The 80m g/(kg·d) group of the new formula was administered orally at 10m L/kg daily from 9:00 to 10:00 am. The normal control group and the high-fat model group were given an equal volume of distilled water. In addition to the normal control group, the remaining groups of rats were fed with a fat emulsion at a volume of 10 ml/kg at a time of 1:00 to 2:00 PM for 28 days. On the 29th day, serum total cholesterol, triacylglycerol and high-density lipoprotein cholesterol, serum apolipoprotein AI, and serum apolipoprotein B were measured in each group. At the same time, livers of each group were taken for pathological examination to observe the degree of hepatic steatosis. MAIN OUTCOME MEASURES: Lipids and hepatic steatosis in the rats after modeling. RESULTS: Sixty rats were used in the experiment and all the results were analyzed. 1Comparison of blood lipid status in each group after modeling: Serum total cholesterol, low-density lipoprotein cholesterol and high-density lipoprotein cholesterol were significantly increased in the high-fat model group, which was significantly different from the normal control group (P < 0.01). Serum triacylglycerol only slightly increased (P > 0.05); Ginkgo biloba extract new formula 20 ~ 80m g / (kg · d) group can inhibit the formation of hyperlipidemia in rats, showing: Serum total cholesterol, triacylglycerol, and low-density lipoprotein cholesterol were significantly lower (P < 0.05 or 0.01), and there was a certain increase in high-density lipoprotein cholesterol, serum apolipoprotein AI, and serum apolipoprotein B reduction. Among them, the new formulation of Ginkgo biloba extract 80m g/(kg·d) was the most significant (P < 0.05). 2Comparison of hepatic steatosis in rats of each group after modeling: liver pathological examination of rats in high-fat model group shows diffuse fat vacuoles; new formula of Ginkgo biloba extract 40,80m g/(kg·d) group The degree of hepatic steatosis in rats has been reduced to a certain extent, especially in the 80m g/(kg·d) group of new formula of Ginkgo biloba extract. Conclusion: It is suggested that the new formula of Ginkgo biloba extract can effectively correct the lipid metabolism of hyperlipidemia and has a good preventive effect on hyperlipidemia caused by high fat diet.
