目的:建立鉴定半夏、水半夏的分子标记方法。方法:利用22条随机引物,对四川两种半夏、广西两种水半夏基因DNA进行RAPD扩增,并对其差异条带进行测序、标记构建探针和Southern杂交。结果:半夏、水半夏基因DNA之间扩增的差异条带较多。通过测序、标记构建探针和杂交确认一半夏特异的标记分子。结论:通过RAPD和构建特异DNA探针能准确鉴别半夏和水半夏。
Objective: To establish a molecular marker method to identify Pinellia ternate and Pinellia ternate. METHODS: Twenty-two random primers were used to amplify RAPD gene DNA from two species of Pinellia and Guangxi, and the differential bands were sequenced, labeled and probed for Southern hybridization. Results: There were more differential bands amplified between Pinellia ternate and Pinellia genus DNA. Half of the Xia-specific marker molecules were identified by sequencing, constructing probes and hybridization. CONCLUSION: Pinellia ternata and Pinellia ternate can be accurately identified by RAPD and the construction of specific DNA probes.
