用不同浓度配比的生长素和细胞分裂素诱导梨枣叶片和茎段愈伤组织的产生,并研究了不定芽诱导的最佳配方,建立了梨枣叶片和茎段的再生体系。结果表明,梨枣叶片愈伤组织诱导的最佳培养基为M S+2,4-D 1.5m g.L-1+6-BA 0.5 m g.L-1;茎段为M S+2,4-D 1.0 m g.L-1+6-BA 0.5 m g.L-1。叶片不定芽诱导的最佳培养基为M S+IBA 0.1 m g.L-1+6-BA 1.5 m g.L-1。A gNO3能阻止叶片外植体褐化并有效地促进叶片愈伤组织分化。茎段能在同一培养基上产生愈伤组织并直接分化出不定芽。
The different concentrations of auxin and cytokinin were used to induce the callus formation of the leaves and stem segments of Lycium barbarum L. The optimal formulation induced by adventitious buds was studied and the regeneration system of the leaves and stem segments was established. The results showed that the optimum medium for the callus induction of L. japonica leaves was M S + 2,4-D 1.5 m gL-1 + 6-BA 0.5 m gL-1; 1.0 m gL-1 + 6-BA 0.5 m gL-1. The optimal medium for adventitious bud induction was M s + IBA 0.1 m g · L-1 + 6-BA 1.5 m g · L-1. A gNO3 prevented leaf explants from browning and effectively promoted leaf callus differentiation. Stem segments can produce callus on the same medium and differentiate adventitious buds directly.
