目的:检测中药复方通络救脑及其主要成分三七总皂苷和京尼平苷对神经突触可塑性有无促进作用。方法:实验分为对照组和给药组,采取细胞活力检测法,即CCK-8检测法检测药物对稳转的过表达的瑞典型突变淀粉前体蛋白(swAPP)突变的人神经母细胞瘤(SY5Y)细胞的增殖曲线的影响;通过Western blotting检测药物对于神经细胞突触相关蛋白树突棘肌动蛋白结合蛋白(drebrin)和突触后密度蛋白-95(PSD-95)的表达的变化;用神经生长因子(NGF)诱导PC12细胞长出神经突起后,检测药物对于细胞突起的作用。结果:复方通络救脑及其主要组成成分京尼平苷和三七总皂苷不影响过表达的瑞典型突变淀粉样前体蛋白的神经母细胞瘤(swAPP-SY5Y)的细胞增殖曲线,且复方通络救脑中京尼平苷能够显著增加swAPP-SY5Y细胞drebrin和PSD-95的表达(P<0.01),显著增加PC12细胞总突起数(P<0.01);而三七总皂苷没有相应的作用。结论:复方通络救脑能促进神经细胞的突触可塑性,初步断定起主要作用的成分是京尼平苷而不是三七总皂苷。
OBJECTIVE: To detect the effect of Tonglianjiunao, a traditional Chinese medicine compound, and its main component Panax notoginseng saponins and geniposide on the synaptic plasticity. Methods: The experiment was divided into the control group and the administration group. The cell viability assay (CCK-8) was used to test the effect of the drug on the steady-staged overexpression of the swedish mutant amylin precursor protein (swAPP) mutant human neuroblastoma (SY5Y) cells. Western blotting was used to detect the changes of the expression of drebrin and PSD-95 in neurons synaptic-related protein dendritic spines ; PC12 cells were induced to grow neurite by nerve growth factor (NGF), and the effect of drugs on cell protrusion was detected. Results: Compound Tongluo to save the brain and its main components of geniposide and notoginsenoside did not affect the cell proliferation curve of overexpressed Swedish mutant amyloid precursor protein neuroblastoma (swAPP-SY5Y), and Compound melatonin to save brain in geniposide can significantly increase the expression of drebrin and PSD-95 in swAPP-SY5Y cells (P <0.01), and significantly increase the total number of PC12 cells (P <0.01); while there is no corresponding effect. Conclusion: Compound Tongluojiuao can promote the synaptic plasticity of nerve cells. It is initially concluded that geniposide rather than Panax notoginseng saponin is the main component.
