目的:对我国云南省边境线获得的未知病毒进行分离鉴定。方法:将样本进行随机聚合酶链式反应,纯化链接到载体中进行序列的测定,确定病毒为甲病毒属类盖他病毒,然后用特异性引物进行衣壳蛋白基因和3’UTR区序列进行DNA测序,分析比较该病毒的分子生物学遗传特征,绘制系统发育树。结果:该病毒为盖塔病毒。结论:该病毒与韩国分离株和河北两株亲缘关系较近,而与MM2021马来西亚原始毒株亲缘关系较远。
OBJECTIVE: To isolate and identify unknown viruses obtained from the border of Yunnan Province in China. METHODS: The samples were subjected to random polymerase chain reaction (PCR), and the sequence was determined by PCR. The virus was identified as alphavirus and then the capsid protein gene and 3’UTR region were sequenced using specific primers DNA sequencing, analysis and comparison of the molecular genetic characteristics of the virus, mapping phylogenetic tree. Results: The virus is Geeta virus. Conclusion: The virus is closely related to two isolates from Korea and Hebei, but has a close genetic relationship with the original strain MM2021 in Malaysia.
