Continuous exposure of breast cancer cells to adriamycin(Adr) induces high expression of P-gp and multiple drug resistance.However, the biochemical process and the underlying mechanisms for the gradually induced resistance are not clear.GC/MS profiling and PCA analysis of the data revealed that Adr significantly induced a reprogrammed metabolism, e.g., the markedly depressed biosynthesis of proteins, purines, pyrimidines, glutathione, and glycolysis, while enhanced glycerol metabolism of MCF7S cells.The reduced synthesis of glutathione suggested the weakened capacity for anti-oxidative stress, while the elevated glycerol metabolism indicated enhanced generation of ROS.It was interesting to find that Adr stimulated higher ROS and Pgp expression, where scavenger N-acetylcysteine could reverse the up-regulation of P-gp.
