  鲨鱼单域抗体(single-domain antibodies,sdAbs)是目前已知最小且能与抗原发生特异性结合活性的抗体分子,其分子量低至12kDa左右。因其具有分子量小、溶解能力强等特点,可以穿过血脑屏障,更有效的与抗原结合。鲨鱼单域抗体的FR1与CDR3和CDR3与FR3之间还存在两对非典型的二硫键,因此其作为药物能更加稳定地存在于机体内,更好发挥作用。

条纹斑竹鲨是一种浅海底栖小型鲨鱼,能够产生免疫球蛋白新抗原受体(Ig New Antigen Receptor IgNAR)其特点是仅由重链组成,是目前已知的尺寸最小的抗体分子。我们通过对鲨鱼脾脏及淋巴细胞进行转录组测序,比对分析获得数条鲨鱼IgNAR序列,针对已知序列设计特异性引物。提取鲨鱼脾脏及淋巴细胞RNA,进行RT-PCR,对鲨鱼抗体进行鉴定。
