稻瘟病菌中Chm1下游相关作用蛋白探究

  稻瘟病菌(Magnaporthe oryzae)是研究植物宿主与病原菌之间相互作用关系的一种模式真菌,其主要侵染水稻,造成大量减产.Chm1是一类丝氨酸/苏氨酸蛋白激酶PAK家族的一员,PAKs通常作为Rho GTP酶的效应蛋白起作用.研究证明,酵母中Chm1同源基因Ste20与Cdc42互作,共同参与调控菌丝的生长和信息素反应等.稻瘟病菌中MoChm1通过一个高度保守的P21结合区域(PBD domain)与小G蛋白Rac1结合,对于调节稻瘟病菌的产孢途径有着重要的作用.因此为了进一步说明产孢途径中Rac1-Chm1这条信号途径,我们利用RNA-sequence和iTRAQ技术对MoChm1缺失突变体进行了研究.结果表明,相对野生型Guy11,有部分基因在MoChm1缺失突变体产孢过程中RNA与蛋白水平产生不同程度的上调与下调,从中筛选出3个上调或下调较明显的基因,分别为MoUbr1(上调8倍)、MoFre(下调16倍)和MoCdc3(上调2倍),并对其逐一进行敲除.表型分析结果显示,相较于Guy11,MoUbr1和MoFre敲除突变体在生长、产孢和致病性等表型上均没有明显的差异,但MoCdc3敲除突变体生长速度下降,产孢量极低,出现单隔膜和无隔膜的异常分生孢子,而以上这些表型都与MoChm1缺失突变体类似.通过Pull Down验证,MoCdc3与MoChm1存在互作关系,因此MoCdc3极有可能是MoChm1假定的下游靶标.
