目的 :研究不同类型视网膜母细胞瘤 (Retinoblastoma ,Rb)标本中的DNA含量及细胞周期各时相分布 ;探讨影响Rb增殖和分化的因素。方法 :应用流式细胞学技术检测 2 0例Rb标本的DNA含量及细胞周期各时相分布 ,定量分析其增殖指数 ,并将各指标进行比较。结果 :正常视网膜组织细胞与不同分化类型Rb的DNA组方图明显不同。Rb的DNA指数 (DI)及S期细胞百分比 (SPF)在不同的Rb视神经侵犯程度、分化类型中具有显著的统计学差异 :Rb累及视神经筛板后的Rb组的DI和SPF值均高于未累及筛板组 (P <0 0 5 ) ;分化型Rb的DI明显低于未分化型组 (P <0 0 1) ,而SPF明显高于未分化组 (P<0 0 1)。结论 :视网膜母细胞瘤DNA含量的研究有助于认识Rb的发生、发展规律 ,具有一定的理论价值与应用前景
OBJECTIVE: To study the DNA content and cell cycle distribution in different types of retinoblastoma (Rb) specimens and to explore the factors affecting the proliferation and differentiation of Rb. Methods: DNA content and cell cycle distributions of 20 Rb specimens were detected by flow cytometry. The proliferation index was quantitatively analyzed and the indexes were compared. Results: The histogram of DNA in normal retinal tissue cells and Rb with different differentiation types was significantly different. The DNA index (DI) and S phase cell percentage (SPF) of Rb were significantly different among different types of Rb optic nerve invasion and differentiation type. The DI and SPF values of Rb group and Rb group (P <0.05). The DI of differentiated Rb was significantly lower than that of undifferentiated group (P <0.01), while the SPF of differentiated Rb was significantly higher than that of undifferentiated group (P <0.01). Conclusion: The study on the DNA content of retinoblastoma is helpful to understand the occurrence and development of Rb, and has certain theoretical value and application prospect
