目的:观察模拟失重和辐射对大鼠脾脏T淋巴细胞功能和胸腺细胞凋亡的影响,及中药太空燮理汤的调节作用。方法:大鼠随机分为正常对照、悬吊、悬吊加辐射、中药共4组。后3组大鼠头低位-30度尾部悬吊连续16 d,其中悬吊加辐射组和中药组大鼠于悬吊第8天接受单次总量为4.5Gy的60Co-γ射线照射,中药组大鼠从实验第1天开始按7g.kg-1.d-1给予中药太空燮理汤灌胃,其余各组大鼠灌服等容积的生理盐水。实验第16天处杀各组大鼠,采用MTT比色法、放免法检测脾T淋巴细胞增殖率和分泌IL-2水平,利用流式细胞仪检测胸腺细胞凋亡。结果:较之正常对照组,悬吊组和悬吊加辐射组大鼠脾淋巴细胞增殖率和分泌IL-2水平显著降低(P<0.01),胸腺细胞凋亡率显著升高(P<0.05);悬吊组和悬吊加辐射组之间无显著差异(P>0.05);较之于悬吊加辐射组,中药组大鼠脾淋巴细胞增殖率显著增加(P<0.05),T淋巴细胞分泌IL-2水平呈增加趋势(P>0.05),胸腺细胞凋亡率显著降低(P<0.01)。结论:单纯失重和失重加辐射均可引起机体淋巴细胞功能降低,太空燮理汤对失重加辐射条件下的机体免疫功能具有调节作用。
OBJECTIVE: To observe the effects of simulated weightlessness and radiation on rat spleen T-lymphocyte function and apoptosis of thymocytes, and the regulatory effect of Chinese herbal medicine Wei Li Tang Li Tang. Methods: Rats were randomly divided into four groups: normal control, suspension, suspension plus radiation, and traditional Chinese medicine. Rats in the last 3 groups were suspended at the head of -30 degrees for 16 consecutive days. Rats in the suspension plus radiation group and the traditional Chinese medicine group received a single dose of 60G-γ rays with a total of 4.5 Gy on the 8th day of suspension. The rats in the group were given intragastric administration of 7g.kg-1.d-1 gavage from the first day of the experiment, and rats in the other groups were given an equal volume of normal saline. Rats in each group were killed on the 16th day of the experiment. MTT assay and radioimmunoassay were used to detect the proliferation rate of spleen T lymphocytes and the secretion of IL-2. Flow cytometry was used to detect apoptosis of thymocytes. RESULTS: Compared with the normal control group, the spleen lymphocyte proliferation rate and secretion of IL-2 were significantly decreased (P<0.01), and the thymocyte apoptosis rate was significantly increased in the suspension group and suspension plus irradiation group (P<0.05). There was no significant difference between the suspension group and the suspension plus radiation group (P>0.05). Compared with the suspension plus radiation group, the spleen lymphocyte proliferation rate of the Chinese herb group was significantly increased (P<0.05). The level of IL-2 secreted by cells increased (P>0.05), and the apoptosis rate of thymocytes decreased significantly (P<0.01). Conclusion: Simple weightlessness, weightlessness plus radiation can all cause the decrease of lymphocyte function of the body. Space Qi Li Tang can regulate the immune function under weightlessness and radiation conditions.
