目的观察葛根芩连结肠定位(GGQLJC)片对湿热内蕴型溃疡性结肠炎(UC)家兔模型Th1/Th2细胞因子平衡的影响,探讨其治疗湿热内蕴型UC的免疫学机制。方法采用高脂高糖饮食法造出家兔湿热内蕴型体质,结合三硝基苯磺酸(TNBS)法建立湿热内蕴型UC模型;将家兔分为7组,每组8只:正常组,模型组,GGQLJC片高剂量组,GGQLJC片中剂量组,GGQLJC片低剂量组,阳性西药柳氮磺胺吡啶(SASP)组,传统葛根芩连(GGQL)片组;对家兔进行临床活动度观察、疾病活动度指数(DAI)评分、结肠黏膜损伤指数(CMDI)评分、组织学损伤(TDI)评估,检测结肠长度重量、脾脏和胸腺的重量、结肠组织中髓过氧化物酶MPO活性、采用酶联免疫吸附法(ELISA)分析家兔血清和结肠组织中Th1细胞因子IL-1β、TNF-α和Th2细胞因子IL-4、IL-13水平。结果各治疗组能够有效缓解湿热内蕴型UC家兔的临床症状,降低DAI评分、CMDI评分、TDI评分,增加结肠长度、降低结肠和脾脏重量、增加胸腺的重量,降低MPO活性,降低家兔血清及结肠组织Th1细胞因子IL-1β和TNF-α并提高Th2细胞因子IL-4、IL-13的表达,其中以GGQLJC片高剂量组及SASP组作用效果更为显著。结论 GGQLJC片可以改善湿热型溃疡性结肠炎的炎症反应,调节Th1/Th2细胞的平衡,这可能是其治疗湿热内蕴型UC的作用机制之一。
Objective To observe the effect of GeGZJG tablet on the Th1 / Th2 cytokine balance in a rabbit model of damp-heat intrinsic ulcerative colitis (UC), and to explore its immunological mechanism for the treatment of endogenous damp - heat type UC. The rabbits were divided into 7 groups with 8 in each group: The normal group, the model group, the GGQLJC tablet high dose group, the GGQLJC tablet middle dose group, the GGQLJC tablet low dose group, the positive western medicine SASP group, the traditional Gegen Qinlian group (GGQL) Activity observation, DAI score, CMDI score, histological damage (TDI) assessment, length of colon, weight of spleen and thymus, myeloperoxidase MPO The Th1 cytokines IL-1β, TNF-α and Th2 cytokines IL-4 and IL-13 in rabbit serum and colon tissue were detected by enzyme-linked immunosorbent assay (ELISA) Results The treatment groups were able to effectively alleviate the clinical symptoms of damp-heat intrinsic UC rabbits, reduce the DAI score, CMDI score, TDI score, increase the length of the colon, reduce the weight of the colon and spleen, increase the weight of the thymus, reduce the activity of MPO, Serum and colon tissue Th1 cytokines IL-1β and TNF-α and increase the expression of Th2 cytokines IL-4, IL-13, of which GGQLJC tablets high-dose group and SASP group effect more pronounced. Conclusion GGQLJC tablet can improve the inflammatory response of damp-heat ulcerative colitis and regulate the balance of Th1 / Th2 cells, which may be one of its mechanisms of treatment of damp-heat intrinsic UC.
