神经毒素MPTP可使人和某些动物产生典型的帕金森样症状。本实验以恒河猴为实验对象,研究~3H-MPTP在猴脑内的分布趋势,探讨帕金森病的发病机理。 材料方法 取恒河猴与大鼠全脑,低温速冻,连续冠状面切片。用5~300nM各种浓度的~3H-MMP溶液对大鼠脑冰冻切片孵育,漂洗后测量鼠脑切片的放射性记数,绘制成~3H-MPTP与脑组织结合的饱和曲线,并以200nM定为~3H-MPTP与猴脑结合的饱和浓度。取猴脑四
Neurotoxin MPTP produces typical Parkinson-like symptoms in humans and certain animals. In this study, Rhesus macaques were used as experimental subjects to study the distribution of ~ 3H-MPTP in the monkey brain and to explore the pathogenesis of Parkinson’s disease. Material Methods Rhesus monkey and rat whole brain, frozen at low temperature, continuous coronal section. The rat brain sections were incubated with various concentrations of ~ 3H-MMP at concentrations ranging from 5 to 300 nM, and radioactive counts of brain sections were measured after rinsing.
