目的:建立HPLC测定穿黄清热胶囊中穿心莲内酯和脱水穿心莲内酯含量的方法。方法:色谱柱为Diamonsil C18(2)柱(4.6 mm×250 mm,5μm),以甲醇-水(45∶55)为流动相,检测波长254 nm。结果:穿心莲内酯与脱水穿心莲内酯的线性范围内分别为0.027 8μg～1.39μg和0.061 4μg～3.07μg,相关系数r均为0.999 9;回收率分别为100.43%(RSD 1.98%)和98.27%(RSD 1.58%)。结论:本方法稳定、重现性好,可作为本制剂的含量测定方法。
OBJECTIVE: To establish a HPLC method for the determination of andrographolide and dehydroandrographolide in Chuan Huang Qing Re Heat Capsule. Methods: Diamonsil C18 (2) column (4.6 mm × 250 mm, 5 μm) was used as the mobile phase. The mobile phase consisted of methanol and water (45:55). The detection wavelength was 254 nm. Results: The linear range of andrographolide and dehydroandrographolide were 0.027 8μg ~ 1.39μg and 0.061 4μg ~ 3.07μg, the correlation coefficient r was 0.999 9; the recoveries were 100.43% (RSD 1.98%) and 98.27% (RSD 1.58%). Conclusion: The method is stable, reproducible, and can be used as the method for the determination of the content of this preparation.
