目的 观察昆明山海棠 (TH)总生物碱诱导白血病细胞凋亡的发生情况。方法 应用台盼蓝染色法 ,荧光双重染色法 ,DNA电泳结合流式细胞仪对HL 6 0细胞的凋亡进行观测。结果 HL 6 0细胞在TH总生物碱作用下出现典型的细胞凋亡特征 ,包括细胞形态改变、DNA梯状带及亚G1峰 ,且动态变化过程较为一致。结论 TH总生物碱具有较强的诱导白血病HL 6 0细胞凋亡的能力 ,总生物碱是TH诱导凋亡的有效成份之一。
Objective To observe the occurrence of apoptosis in leukemic cells induced by total alkaloids in Kunming Mountain jellyfish (TH). Methods Trypan blue staining, fluorescence double staining, DNA electrophoresis and flow cytometry were used to observe the apoptosis of HL 60 cells. Results HL 60 cells exhibited typical cell apoptosis characteristics under the action of TH total alkaloids, including changes in cell morphology, DNA ladders, and sub-G1 peaks, and the dynamic changes were consistent. Conclusion TH total alkaloids have a strong ability to induce apoptosis of leukemia HL 60 cells. Total alkaloids are one of the effective components of TH-induced apoptosis.
