目的探索SPRY2分子在肿瘤坏死因子α(TNF-α)诱导的L929细胞毒作用中的调节作用。方法应用RT-PCR方法观察SPRY2基因在不同质量浓度TNF-α刺激作用下表达水平的变化;应用重组腺病毒介导SPRY2基因过表达方法观察其对TNF-α细胞毒活性的调解效应。结果 L929细胞中存在SPRY2基因的天然表达,并且其表达水平随TNF-α刺激质量浓度的升高而升高,过表达SPRY2基因促进TNF-α诱导的L929细胞死亡。结论 SPRY2分子在TNF-α介导的肿瘤细胞杀伤效应中发挥正向调节作用。
Objective To investigate the regulatory role of SPRY2 in tumor necrosis factor α (TNF-α) -induced L929 cytotoxicity. Methods The expression of SPRY2 gene was detected by RT-PCR. The effect of SPRY2 gene on the cytotoxicity of TNF-α was observed by the recombinant adenovirus-mediated SPRY2 gene overexpression. Results The natural expression of SPRY2 gene existed in L929 cells, and the expression level of SPRY2 gene increased with the increase of TNF-α stimulation level. Overexpression of SPRY2 gene promoted TNF-α-induced L929 cell death. Conclusions SPRY2 plays a positive regulatory role in TNF-α-mediated tumor cell killing.
