目的探讨消痰散结方对MKN-45人胃癌裸鼠原位移植瘤微卫星不稳定的作用机制。方法采用MKN-45人胃癌细胞株建立裸鼠皮下瘤模型皮下传3代作为实验模型的瘤源,采用OB胶黏合法建立人胃癌裸鼠原位移植瘤模型。20只模型裸鼠分为模型组和中药组。中药组予消痰散结方干预6周后,测瘤质量,计算抑瘤率并检测5个微卫星不稳定位点(D17S250、D2S123、D5S346、Bat-25和Bat-26)。结果中药组的瘤质量明显低于模型组(P<0.01),其抑瘤率为40.84%。模型组的肿瘤组织中能扩增到5个微卫星位点,其中微卫星位点D2S123、D5S346出现某些延长和缩短。使用消痰散结方干预6周后,微卫星位点D2S123、D5S346的变异趋于稳定。结论消痰散结方能抑制MKN-45人胃癌裸鼠原位移植瘤的微卫星不稳定。
Objective To investigate the mechanism of Xiaotan Sanjie prescription on microsatellite instability of orthotopic transplanted tumor of MKN-45 human gastric cancer in nude mice. Methods The MKN-45 human gastric cancer cell line was used to establish the subcutaneous tumor-bearing model in nude mice and the third generation was used as the tumor source. The in situ xenograft model of human gastric cancer was established by OB glue. 20 nude mice were divided into model group and TCM group. The Chinese medicine group was treated with Xiaotan Sanjie Decoction for 6 weeks. The tumor mass was measured. The tumor inhibition rate was calculated and 5 microsatellite instability sites (D17S250, D2S123, D5S346, Bat-25 and Bat-26) were detected. Results The tumor mass of the traditional Chinese medicine group was significantly lower than that of the model group (P <0.01), and the tumor inhibition rate was 40.84%. In the model group, five microsatellite loci were amplified in the tumor tissues, in which the microsatellite loci D2S123 and D5S346 showed some elongation and shortening. After 6 weeks of intervention with Xiaotan Sanjie Formula, the variation of D2S123 and D5S346 in microsatellite loci tended to be stable. Conclusion Xiaotan Sanjie can inhibit microsatellite instability of orthotopic transplanted tumor of MKN-45 human gastric cancer in nude mice.
