目的:观察滋阴泻火方对达那唑诱导的性早熟大鼠下丘脑GnRH mRNA和Leptin mRNA表达的影响。方法:取SD雌性大鼠60只,随机分为正常对照组、性早熟模型组、曲普瑞林组、中药大剂量组、中药中剂量组、中药小剂量组6组,分别用生理盐水,曲普瑞林,中药高、中、低剂量进行干预,观察滋阴泻火方对性早熟大鼠下丘脑GnRH mRNA和Leptin mRNA表达的影响。结果:经阴道涂片检查,模型组大鼠可观察到规则的性周期出现,持续时间也大致相同。与性早熟模型组比较,曲普瑞林组、中药大剂量组、中药中剂量组下丘脑GnRH mRNA表达显著降低(P<0.05),曲普瑞林组、中药大剂量组下丘脑Leptin mRNA表达显著降低(P<0.05),中药小剂量组未见统计学差异(P>0.05)。结论:滋阴泻火方大剂量可以下调性早熟模型大鼠下丘脑GnRH mRNA和Leptin mRNA表达,能有效抑制Leptin的分泌,进而抑制性激素的释放。
Objective: To observe the effect of Ziyin Xiehuo prescription on the expression of GnRH mRNA and Leptin mRNA in hypothalamus of danazol induced precociousness rats. Methods: Sixty female SD rats were randomly divided into normal control group, precocious puberty model group, triptorelin group, high-dose traditional Chinese medicine group, middle-dose traditional Chinese medicine group and low-dose traditional Chinese medicine group. Six groups were treated with normal saline, Triptorelin, high, medium and low dose of traditional Chinese medicine to observe the effect of Ziyin Xiehuo prescription on hypothalamus GnRH mRNA and Leptin mRNA expression in precocious puberty rats. Results: According to the vaginal smear examination, regular sexual cycles were observed in the model group, with similar durations. Compared with the precocious puberty model group, the expression of GnRH mRNA in the hypothalamus of the triptorelin group, the traditional Chinese medicine middle-dose group and the traditional Chinese medicine middle-dose group was significantly decreased (P <0.05). The levels of Leptin mRNA in the hypothalamus of the triptorelin group and the traditional Chinese medicine high- (P <0.05). There was no significant difference in the low dose of traditional Chinese medicine (P> 0.05). Conclusion: The high dose of Ziyin Xiehu Recipe can down regulate the expression of GnRH mRNA and Leptin mRNA in the hypothalamus of precocious puberty model rats, which can effectively inhibit the secretion of Leptin and then inhibit the release of sex hormones.
