目的 通过大鼠同种异体心脏异位移植 ,探讨胸腺内注射异基因脾细胞对移植心的影响。方法 用供体脾细胞 (SC)在心脏移植前 2 1d预处理受体鼠 ,术后 1周每日腹腔注射环孢素A(CsA) 10mg/kg。检测移植心存活时间、混合淋巴细胞反应 (MLR)及细胞介导的细胞毒试验 (CML)。结果 胸腺注射脾细胞抗原联合短期应用CsA组的移植心平均存活时间较其他组显著延长 (P <0 .0 5 ) ,移植心耐受的受体鼠中MLR增殖活跃 ,产生免疫耐受的受体鼠的T细胞对供体鼠的靶细胞无明显的细胞毒作用。结论 胸腺内注射供体脾细胞抗原预处理受体同时联合短期应用CsA ,可诱导产生移植心脏的免疫耐受 ,且诱导移植耐受的抗原量以 2 .5× 10 7脾细胞为好
Objective To investigate the effect of intrathymic injection of allogeneic spleen cells on cardiac allograft via rat heterotopic cardiac allografts. Methods Recipient rats were pretreated with donor splenocytes (SC) 21 days prior to cardiac transplantation. Cyclosporine A (CsA) 10 mg / kg was intraperitoneally injected one week after the operation. The graft survival time, mixed lymphocyte reaction (MLR) and cell-mediated cytotoxicity (CML) were measured. Results The average survival time of thymus injection of splenocyte antigen combined with CsA group was significantly longer than that of other groups (P <0.05). The multiplication of MLR in recipient mice tolerated by thymus was more active than that of other groups T-cells of the rat do not show any significant cytotoxic effects on the target cells of the donor mice. Conclusion Intrathymic injection of donor splenocyte preconditioning receptor combined with short-term administration of CsA induces immune tolerance of transplanted heart, and the amount of antigen-tolerant transplants is 2.5 × 10 7 spleen cells
