目的:探讨B7-1基因修饰的肿瘤细胞作为瘤苗对小鼠胃癌的治疗效果。方法:在已建立高表达B7-1分子的小鼠前胃癌细胞系MFC-B7的基础上,观察MFC-B7细胞体外激活脾淋巴细胞的能力,体内成瘤性变化及作为瘤苗对荷瘤鼠胃癌的治疗效果。结果:MFC-B7细胞在体外能有效诱导脾淋巴细胞的活化增殖和特异性CTL(CytotoxicTlymphocytes)活性的产生,在体内的成瘤能力明显下降,作为瘤苗接种对荷瘤鼠胃癌有较好的治疗效果。结论:B7-1基因修饰的肿瘤细胞免疫原性明显增强,有希望作为瘤苗用于此类肿瘤的治疗。
Objective: To investigate the effect of B7-1 gene-modified tumor cells as tumor vaccines on gastric cancer in mice. METHODS: On the basis of the establishment of a mouse gastric cancer cell line MFC-B7 with high expression of B7-1 molecule, the ability of MFC-B7 cells to activate splenic lymphocytes in vitro was investigated. The tumorigenicity in vivo was changed and used as a tumor vaccine to tumor-bearing tumors. Rat gastric cancer treatment effect. RESULTS: MFC-B7 cells could effectively induce the proliferation of spleen lymphocytes and the production of specific CTL (CytotoxicTlymphocytes) activity in vitro.
