几家报道表明使用反义寡核苷酸可抑制c-myc基因表达,但均需高浓度的游离寡核苷酸。为了降低寡核苷酸浓度并稳定其对c-myc的反义效应,作者将与c-myc mRNA起始码及其后的4个密码子互补的c-myc反义寡核苷酸与聚-L-赖氨酸(PLL)共价结合并加入肝素(100μg/ml)形成三元复合物。
Several reports indicate that the use of antisense oligonucleotides inhibits c-myc gene expression, but requires high concentrations of free oligonucleotides. To reduce the concentration of oligonucleotides and stabilize their antisense effects on c-myc, the authors compared c-myc antisense oligonucleotides complementary to the c-myc mRNA start codon and the last four codons with poly -L-lysine (PLL) and heparin (100 μg / ml) was added to form a ternary complex.
