本文探讨用~(51)Cr释放试验检测ADCC中影响靶细胞标记及自然释放的各种因素。实验表明标记率随着~(51)Cr原液比放射性的增高以及放射性强度的增高而增高。为保证一定的标记率并避免自然释放过高,选用100微居里的放射性强度,其铬含量不超过10微克为宜。标记细胞存放时间不宜超过一周,且应防止污染。如在自然释放管中加入未标记的CRBG可降低自然释放率。因此,控制好影响自然释放的因素,可使检测ADCC的~(51)Cr释放试验得到较为可靠和满意的结果。
In this paper, ~ (51) Cr release assay was used to detect various factors affecting target cell markers and natural release in ADCC. Experiments show that the labeling rate increases with the radioactivity of ~ (51) Cr solution and the increase of radioactive intensity. To ensure a certain labeling rate and to avoid excessive natural release, the use of radioactive intensity of 100 micro-Curie, the chromium content of not more than 10 micrograms is appropriate. Label cells should not be stored for more than a week, and should prevent contamination. If natural release tube unlabeled CRBG can reduce the natural rate of release. Therefore, controlling the factors that affect the natural release can make the ~ (51) Cr release test of ADCC obtain more reliable and satisfactory results.
