建立了超高效亲水作用色谱-蒸发光散射(UP-HILIC-ELSD)检测牛磺酸颗粒和牛磺酸滴眼液中牛磺酸含量的方法。样品采用乙腈-水稀释,在超高效酰胺基(Amide)亲水作用色谱柱中分离,乙腈-水为流动相,梯度程序洗脱,蒸发光检测器测定。牛磺酸绝对质量在2.5~100 ng时浓度与峰面积响应呈良好的线性关系,相关系数(r)为0.9998,方法定量限为5 ng,回收率在98.9%~100.3%之间,相对标准偏差(RSD)小于2.1%。与反相-液相色谱法对比测定了市售药品及保健品中牛磺酸的含量,表明本方法能准确、快速地测定批量样品中牛磺酸的含量。
A method for the determination of taurine in taurine granules and taurine eye drops by UP-HILIC-ELSD was established. Samples were diluted with acetonitrile-water, separated on Amide hydrophilic columns, acetonitrile-water as mobile phase, gradient elution and evaporated light detector. When the absolute mass of taurine was 2.5-100 ng, there was a good linear relationship between the concentration and the peak area, the correlation coefficient (r) was 0.9998, the limit of quantification was 5 ng and the recovery was between 98.9% and 100.3% Deviation (RSD) is less than 2.1%. Compared with reversed-phase liquid chromatography, the content of taurine in commercial drugs and health products was measured, which showed that the method could accurately and quickly determine the content of taurine in bulk samples.
