用弓形虫可溶性抗原免疫BALB/c 小鼠,取脾细胞,与Sp2/0 细胞融合,获得3 株能稳定分泌抗弓形虫单克隆抗体的细胞。细胞培养上清液和腹水中的单克隆抗体效价分别为1∶16 ~256 和10 - 6 ~10 - 7 。单抗类型均为IgM。杂交瘤细胞经液氮冻存12 个月后,复苏生长良好,分泌活性稳定。在McAb- Dot- ELISA 实验中,所能测出可溶性抗原的最小浓度为10 ng/ mL
BALB / c mice were immunized with soluble antigen of Toxoplasma gondii and splenocytes were obtained and fused with Sp2 / 0 cells to obtain three cells that can stably secrete monoclonal antibody against Toxoplasma gondii. Monoclonal antibody titers in cell culture supernatants and ascites were 1:16 to 256 and 10 - 6 to 10 - 7, respectively. Monoclonal antibody types are IgM. After hybridoma cells were frozen in liquid nitrogen for 12 months, the resuscitated cells grew well and their secretion activity was stable.
