目的观察在高浓度葡萄糖条件下,体外培养的兔视网膜Müller细胞的钙依赖性钾通道的变化。方法高浓度葡萄糖条件下体外培养兔视网膜Müller细胞,应用免疫组织化学技术和透射电镜鉴定Müller细胞,并采用膜片钳技术观察不同时期高糖条件下视网膜Müller细胞钙依赖性钾通道的变化。结果高糖对体外培养的Müller细胞钙依赖性钾通道有阻滞作用,其阻滞作用呈时间依赖性。结论高浓度葡萄糖可能通过阻滞钙依赖性钾通道而降低Müller细胞的生物学功能。
Objective To observe the changes of calcium-dependent potassium channel in cultured rabbit retinal Müller cells under high glucose conditions. Methods Rabbit retinal Müller cells were cultured in vitro with high concentration of glucose. Müller cells were identified by immunohistochemistry and transmission electron microscopy. The changes of calcium-dependent potassium channel in retinal Müller cells were observed by patch-clamp technique. Results High glucose inhibited the calcium-dependent potassium channel of Müller cells cultured in vitro, and its arrest was time-dependent. Conclusion High glucose may reduce the biological function of Müller cells by blocking calcium-dependent potassium channel.
