目的建立赤芝孢子油软胶囊中植物脂肪酸甘油酯的HPLC-ELSD指纹图谱,并与植物油HPLC色谱图进行比较。方法采用高效液相色谱.蒸发光散射检测法(HPLC-ELSD)测定14批赤芝孢子油软胶囊与7种植物油样品。色谱柱为Diamonsil C_(18)柱(250 mm×4.6 mm,5μm),流动相为乙腈-异丙醇(体积比为51:49),流速:1.0 mL·min~(-1),ELSD漂移管温度:47℃,载气流速:3.2 L·min~(-1)。结果建立了赤芝孢子油软胶囊中植物脂肪酸甘油酯的HPLC-ELSD指纹图谱,标定了10个共有色谱峰,采用对照品指认了其中4个峰。赤芝孢子油软胶囊HPLC-ELSD指纹图谱与植物油的液相色谱图存在明显的差异。结论该方法简便、特异、重现性好,可为赤芝孢子油软胶囊的质量控制提供参考。
OBJECTIVE To establish HPLC-ELSD fingerprints of vegetable fatty acid glycerides in Ganoderma lucidum spore oil soft capsule and compare with HPLC chromatogram of vegetable oil. Methods HPLC-ELSD was used to determine 14 batches of Ganoderma lucidum spores oil capsules and 7 vegetable oil samples by high performance liquid chromatography (HPLC). The column was Diamonsil C 18 column (250 mm × 4.6 mm, 5 μm) with a mobile phase of acetonitrile-isopropanol (volume ratio of 51:49), flow rate of 1.0 mL · min -1, ELSD drift Tube temperature: 47 ℃, carrier gas flow rate: 3.2 L · min -1. Results HPLC-ELSD fingerprints of plant fatty acid glycerides were prepared. The common peaks of 10 were calibrated and 4 of them were identified by reference standard. HPLC-ELSD fingerprints of Ganoderma spore oil soft capsule have obvious difference with the liquid chromatogram of vegetable oil. Conclusion The method is simple, specific and reproducible, which can provide reference for the quality control of Ganoderma spore oil soft capsule.
