目的探讨邻苯二甲酸二丁酯(di-n-butyl phthalate,DBP)和邻苯二甲酸二(2-乙基己基)酯(di-2-ethy lhexyl phthalate,DEHP)对雄性大鼠精子生成的联合毒性作用。方法将32只健康清洁级成年SD雄性大鼠按体重随机分为4组,分别为阴性对照组(玉米油)、DBP染毒组(1/20LD50,1.0g/kg)、DEHP染毒组(1/20LD50,1.7g/kg)和DBP+DEHP染毒组(1.0g/kgDBP+1.7g/kgDEHP),每组8只。采用灌胃方式进行染毒,每天1次,每周5d,连续染毒8周。末次染毒24h后,测量大鼠体重、睾丸和附睾及肝脏脏器湿重,测定精子参数(精子数量、精子活动率及精子畸形率),并观察睾丸及附睾组织形态学改变。结果DBP和DEHP联合染毒对大鼠体重增长无交互作用(P>0.05);对睾丸和附睾及肝脏脏器系数存在交互影响,使睾丸和附睾的脏器系数明显降低,肝脏的脏器系数明显升高,表现为协同作用;对精子计数、精子活动率及精子畸形率存在交互影响,使精子数量和存活率明显降低,畸形率明显升高,表现为协同作用。光镜下可见DBP和DEHP联合染毒可引起曲细精管外形不规则、萎缩变性,间质增宽,生精上皮退化变性,甚至几乎消失,生精细胞耗竭,基底部仅有少量的支持细胞且细胞肿胀、变性,出现空泡样改变,基膜尚完整;附睾上皮受损且变薄,间质增宽,管腔内几乎不见成熟精子。结论DBP和DEHP联合染毒对雄性大鼠具有明显的生殖毒性作用,可严重影响精子生成数量和质量,且对附睾有一定的毒作用。
Objective To investigate the effects of di-n-butyl phthalate (DBP) and di-2-ethy lhexyl phthalate (DEHP) on spermatogenesis in male rats Joint toxic effects. Results There was no interaction between DBP and DEHP in rats (P> 0.05). There was interaction between testis, epididymis and liver organ coefficient, and the organ coefficient of testis and epididymis was significantly decreased. The organ coefficient of liver Significantly increased, showing synergies; sperm count, sperm motility and sperm deformity rate of interaction there, the sperm count and survival rate was significantly reduced, deformity rate was significantly increased, showing synergies. Under the light microscope, DBP and DEHP combined exposure can cause irregular shape of seminiferous tubules, atrophic degeneration, interstitial broadening, degeneration of seminiferous epithelium degeneration, even almost disappeared, germinal cell depletion, basal only a small amount of support Cells and cell swelling, degeneration, there vacuolar changes, the basement membrane is still intact; epididymal epithelium damaged and thinning, interstitial broadening, almost no visible mature lumen sperm. Conclusion DBP and DEHP combined with exposure to male rats have significant reproductive toxicity, can seriously affect the quantity and quality of sperm production, and to the epididymis has some toxic effects.
