目的探讨东亚钳蝎蝎毒多肽提取物(PESV)对胰腺癌肝转移的抑制作用。方法筛选出高肝转移的胰腺癌细胞亚型MIA-PaCa2-3,用Transwell小室模型观察PESV对MIA-PaCa2-3细胞体外侵袭能力的影响;尾静脉注射MIA-PaCa2-3细胞建立高肝转移率的裸鼠胰腺癌模型,对比PESV和化疗药物吉西他滨(gemcitabine)以及抗血管生成剂TNP-470对裸鼠胰腺癌肝转移的抑制作用。结果PESV可以抑制MIA-PaCa2-3细胞穿过人工基底膜的能力;PESV处理组肝转移抑制率为70%(7/10),与吉西他滨处理组相近(8/10),明显高于TNP-470处理组(2/9)。结论PESV作为一种“抗血管-细胞毒”药物对胰腺癌肝转移具有明显的抑制作用。
Objective To investigate the inhibitory effect of PESV on hepatic metastasis of pancreatic cancer. Methods The MIA-PaCa2-3 pancreatic cancer cell line with high hepatic metastasis was screened out. The effect of PESV on the invasiveness of MIA-PaCa2-3 cells in vitro was observed by Transwell chamber model. The MIA-PaCa2-3 cells were injected into the tail vein to establish a high liver metastasis Rate of pancreatic cancer in nude mice model compared with PESV and the chemotherapy drug gemcitabine (gemcitabine) and the anti-angiogenic agent TNP-470 on the inhibition of hepatic metastasis of pancreatic cancer in nude mice. Results PESV could inhibit the ability of MIA-PaCa2-3 cells to pass through the artificial basement membrane. The inhibition rate of PESV-induced hepatic metastasis was 70% (7/10), similar to that of gemcitabine-treated group (8/10), significantly higher than that of TNP- 470 treatment group (2/9). Conclusion PESV as a “anti-vascular-cytotoxic” drug has a significant inhibitory effect on hepatic metastasis of pancreatic cancer.
