GC-MS法同时检测南瓜果实肌醇类和单、双糖类物质的研究

为了高效同时测定南瓜果实中的肌醇类和单、双糖类物质,采用气相色谱-质谱(Gas chromatography mass spectrometer,GC-MS)联用技术,以不同提取试剂、不同提取温度和步骤的不同来确定南瓜肌醇的最佳提取方法,并采用对南瓜果实样品进行全谱扫描和选择离子扫描。得到最优方案:南瓜样品在60%甲醇溶液于70℃水浴1 h,真空干燥以后,加水重新溶解后直接进行衍生化,其提取效果最佳;采用GC-MS扫描确定了南瓜果实样品中的主要代谢成分半乳糖、葡萄糖、D-手性肌醇、肌-肌醇、果糖和蔗糖。试验方法简单高效,GC-MS的进样时间短,出峰完全,可同时分析多个物质,重复性好,结果精确稳定。 In order to efficiently and simultaneously determine the myo-inositol and mono- and di-saccharides in pumpkin fruit, the gas chromatography mass spectrometry (GC-MS) To determine the best pumpkin inositol extraction methods, and the pumpkin fruit samples using full-spectrum scanning and selective ion scan. The optimal solution was obtained: the pumpkin sample was subjected to water bathing at 70 ℃ for 1 h in 60% methanol solution and vacuum-dried. The pumpkin sample was directly derivatized with water after redissolution, and its extraction effect was the best. GC- The main metabolites galactose, glucose, D-chiro-inositol, myo-inositol, fructose and sucrose. Test method is simple and efficient, GC-MS injection time is short, the peak is complete, can simultaneously analyze multiple substances, good repeatability, accurate and stable results.
