目的:研究IL-24基因与NCTD联合作用对A549细胞增殖和凋亡的影响。方法:IL-24和(或)NCTD干预A549细胞24 h后,MTT法检测A549细胞增殖抑制率;用流式细胞仪检测细胞凋亡及周期的变化;Western blot法检测Bax、Bcl-2、VEGF和HSP-70蛋白表达水平。结果:IL-24联合NCTD作用24 h对A549细胞增殖抑制率为60.3%,与NCTD/IL-24组比较差异有显著性;IL-24联合NCTD作用对A549细胞凋亡率为32.5%;IL-24和(或)NCTD作用A549细胞后Sub-G1期细胞增多;NCTD/IL-24作用24 h后Bax表达上调,Bcl-2、VEGF表达下调,联合组Bax蛋白表达上调,Bcl-2、VEGF、HSP-70表达下调,尤其VEGF、HSP-70表达下调明显。结论 :IL-24联合NCTD对A549细胞有协同抑制作用。
Objective: To investigate the effect of IL-24 gene and NCTD on the proliferation and apoptosis of A549 cells. Methods: A549 cells were treated with IL-24 and / or NCTD for 24 h. MTT assay was used to detect the proliferation inhibition rate of A549 cells. Flow cytometry was used to detect apoptosis and cycle changes. Western blot was used to detect the expression of Bax, Bcl-2, VEGF and HSP-70 protein expression levels. Results: The inhibitory rate of IL-24 combined with NCTD on A549 cells was 60.3% at 24 h, which was significantly different from that of NCTD / IL-24 group. The apoptosis rate of A549 cells treated with IL-24 and NCTD was 32.5% The expression of Bcl-2 and VEGF was up-regulated in Bcl-2 and Bcl-2 cells after NCTD / IL-24 treatment for 24 h, VEGF, HSP-70 expression was down-regulated, especially VEGF, HSP-70 expression was significantly down-regulated. Conclusion: IL-24 combined with NCTD has a synergistic inhibitory effect on A549 cells.
