目的:测定杠柳毒苷、杠柳次苷、杠柳苷元与人血浆蛋白的结合率。方法:采用平衡透析法结合HPLC,使用Zorbax SB-C18色谱柱(2.1 mm×150 mm,5μm),杠柳毒苷、杠柳次苷、杠柳苷元分别采用乙腈-水(26∶74)、(30∶70)、(28∶72)为流动相,流速0.3 m L·min-1,检测波长220 nm。结果:在低、中、高不同质量浓度下,杠柳毒苷、杠柳次苷、杠柳苷元和人血浆蛋白的平均结合率分别为83.48%、94.19%、84.04%。结论:杠柳毒苷、杠柳次苷、杠柳苷元和人血浆蛋白的结合率大于80%,为高血浆蛋白结合的化合物。
OBJECTIVE: To determine the binding rate of pipaloxine, glucuronide, pipaloside and human plasma protein. METHODS: Perfoside, bariposide and perylene glycoside were separated on a Zorbax SB-C18 column (2.1 mm × 150 mm, 5 μm) using equilibrium dialysis coupled with HPLC using acetonitrile-water (26:74), 30:70), (28:72) as the mobile phase at a flow rate of 0.3 m L · min-1 with a detection wavelength of 220 nm. Results: The average binding rates of pipaloxine, glufosinate, pipalustin and human plasma protein were 83.48%, 94.19% and 84.04% at low, medium and high concentrations, respectively. CONCLUSION: The binding rates of pipaloxine, glufosinate, pipalidin and human plasma proteins are greater than 80% and are high plasma protein-binding compounds.
