目的:观察EB病毒BHRF1反义寡核苷酸片段对鼻咽癌SUNE1细胞株增殖的影响。方法:PCR、3HTdR的掺入法及电泳等。结果:PCR从SUNE1细胞DNA中扩增出EB病毒的4种(BHRF1、DNA酶、胸苷激酶及核抗原1)全基因片段;适当浓度BHRF1反义寡核苷酸片段能引起无血清培养的SUNE1细胞株增殖受抑制;透射电镜及琼脂糖凝胶电泳检测结果发现与SUNE1细胞发生凋亡有关。结论:适当浓度BHRF1反义寡核苷酸片段能抑制无血清培养的SUNE1细胞株的增殖。
Objective: To observe the effect of EBV BHRF1 antisense oligonucleotide fragments on the proliferation of nasopharyngeal carcinoma SUNE 1 cell line. Methods: PCR, 3H TdR incorporation and electrophoresis. Results: Four kinds of whole genome DNA fragments of EB virus (BHRF1, DNase, thymidine kinase and nuclear antigen 1) were amplified by PCR from SUNE1 cells. The appropriate concentration of BHRF1 antisense oligonucleotide fragments could cause no The proliferation of cultured SUNE 1 cell lines was inhibited by transmission electron microscopy and agarose gel electrophoresis test results and SUNE 1 cell apoptosis. Conclusion: The appropriate concentration of BHRF1 antisense oligonucleotide fragments can inhibit the proliferation of serum-free cultured SUNE 1 cell lines.
