目的研究TGF-β、Bmp/Smad信号通路基因在不同证候H22肝癌荷瘤小鼠肾上腺的表达特征。方法采用小鼠标准化四诊及辨证方法,以及GeneChip Mouse Exon1.0STArray等技术,检测H22荷瘤小鼠接种第8天(早期)邪毒壅盛证和气虚证、接种第21天(中期)阳气虚证、接种第29天(中晚期)气阴阳虚证共4个证候肾上腺基因表达的差异。通过系统聚类法分析不同证候肾上腺Smad信号通路基因表达模式的相似性及其差异特征。结果就证候而言,邪毒壅盛证与气虚证的基因表达模式相似,而阳气虚证和气阴阳虚证的基因表达模式类似。就基因而言,TGF-βr1、2、3等基因具有相似的表达模式;Smad2与Smad4,Bmp1与Bmp4也具有相似的表达模式。Smad信号通路中,TGF-β受体及Bmp受体表达量分别高于TGF-β与Bmp基因。结论芯片数据提示,H22荷瘤小鼠肿瘤发生后,不同证候的肾上腺Smad通路基因多数被激活,且基因表达模式可能与不同证候有关。
Objective To investigate the expression characteristics of TGF-β and Bmp/Smad signaling pathway genes in the adrenal glands of H22 tumor bearing mice with different syndromes. METHODS: Four mouse standardization methods and syndrome differentiation methods were used, and techniques such as GeneChip Mouse Exon 1.0STArray were used to detect H22 tumor-bearing mice inoculated on the 8th day (early stage) of cult poisoning and qi deficiency syndrome, and on the 21st day (middle stage) of inoculation. Differences in gene expression of adrenal glands in 4 syndromes of Qi deficiency syndrome and qi-yin-yang deficiency syndrome on the 29th (middle and late) inoculation. Systematic clustering analysis was used to analyze the similarity and difference of gene expression patterns of adrenal glands in different syndromes. As for the results of the syndrome, the pattern of gene expression of the cult poison and qi deficiency syndrome is similar to that of the qi deficiency syndrome, and the gene expression patterns of the yang-qi deficiency syndrome and the qi-yin-yang deficiency syndrome are similar. In terms of genes, TGF-β1, 2, 3, and other genes have similar expression patterns; Smad2 and Smad4, Bmp1 and Bmp4 also have similar expression patterns. In the Smad signaling pathway, the expression levels of TGF-β receptor and Bmp receptor were higher than those of TGF-β and Bmp genes, respectively. Conclusion The data from the microarray data suggest that after tumorigenesis of H22 tumor-bearing mice, the adrenal gland Smad pathway genes of different syndromes are mostly activated, and the gene expression pattern may be related to different syndromes.
