目的:通过分析柴胡药材中的矿质元素与有效成分之间的关系,探究其对柴胡药材质量的影响,为提高栽培种柴胡的药材质量提供参考。方法:采用微波消解法消解,采用电感耦合等离子体原子发射光谱法(ICP-AES)测定24批样品中Fe,Mg等16种矿质元素的含量,根据《中国药典》2015年版标准,利用HPLC测定指标成分柴胡皂苷a,d的含量,流动相为水(A)-乙腈(B)梯度洗脱(0~5 min,35%~40%B;5~15 min,40%~45%B;15~25 min,45%~55%B;25~30 min,55%~90%B;30~45 min,90%~35%B),流速1.0 mL·min~(-1),进样量20μL,柱温25℃,检测波长210 nm。利用SPSS 17.0软件进行双变量相关方法分析24批样品中矿质元素与有效成分之间的相互关系。结果:不同柴胡样品中矿质元素和有效成分含量都有较大差异,有效成分柴胡皂苷a,d的含量与Cu元素相关系数分别为0.349和0.425,呈显著正相关,与Na元素的相关系数分别为-0.517和-0.420,呈显著负相关。结论:柴胡药材中有效成分柴胡皂苷a,d含量与Cu和Na等矿质元素之间的存在一定关系,建议通过外源施入矿质元素的栽培措施来调控柴胡皂苷a,d在柴胡根中合成,为栽培种柴胡质量的提高提供实验依据。
Objective: To analyze the relationship between the mineral elements in Bupleurum medicinal materials and its active ingredients and explore its influence on the quality of Bupleurum medicinal materials, and to provide references for improving the quality of medicinal materials of Bupleurum chinense. Methods: The contents of 16 mineral elements such as Fe and Mg in 24 batches of samples were determined by inductively coupled plasma atomic emission spectrometry (ICP-AES) and digested by microwave digestion. According to the Chinese Pharmacopoeia 2015 edition, the content of 16 mineral elements was determined by HPLC The content of saikosaponin a, d of the indicator components was eluted with a gradient of water (A) -acetonitrile (B) (0-5 min, 35-40% B; 5-15 min, 40-45% B ; 15-25 min, 45-55% B; 25-30 min, 55% -90% B; 30-45 min, 90-35% B) at a flow rate of 1.0 mL · min -1 The sample volume was 20μL, the column temperature was 25 ℃ and the detection wavelength was 210 nm. SPSS 17.0 software was used to perform a bivariate correlation analysis of the correlation between the mineral elements and the active ingredients in 24 batches of samples. Results: The contents of mineral elements and active ingredients in different Bupleurum samples were quite different. The content of Saikosaponin a, d and the correlation coefficient of Cu element were 0.349 and 0.425 respectively, which were positively correlated with Na element The coefficients were -0.517 and -0.420 respectively, showing a significant negative correlation. CONCLUSION: The content of saikosaponin a and d in active ingredients of Bupleurum is related to the existence of mineral elements such as Cu and Na. It is suggested that the cultivation of exogenous mineral elements should be used to control the content of saikosaponin a and d in Chai Hu Genzhong synthesis, for the cultivation of Bupleurum quality to provide experimental evidence.
