目的 了解烟台地区肾综合征出血热 (HFRS)患者血清中IgG、IgM抗体水平 ,确定引起该地区HFRS流行的汉坦病毒的型别分布。方法 收集临床HFRS急性期和恢复期患者血清 ;用ELISA检测IgG、IgM抗体 ;用交叉空斑减少中和试验检测中和抗体 ;采用Trizol法提取患者血清中HFRS病毒RNA ,用套式PCR产物做TA试验 ,测定核苷酸序列。结果 HFRS患者血清IgM阳性率为82 2 % (88 10 7) ,IgG阳性率为 85 7% (6 6 77)。该地区两城市 38份HFRS患者血清中 ,有 32份属家鼠型病毒 (SEO)感染 ,另 6份未能定型 ;另一城市 16份HFRS患者血清中 ,有 15份属姬鼠型病毒感染 ,1份未定型。该地区HFRS病毒与SEO的同源性达 90 %以上。结论 引起烟台地区HFRS流行的汉坦病毒 ,属于以SEO为主的混合型病毒。
Objective To understand the serum levels of IgG and IgM antibodies in patients with hemorrhagic fever with renal syndrome (HFRS) in Yantai area and to determine the type distribution of Hantaan virus that causes the epidemic of HFRS in this area. Methods Serum samples were collected from clinical HFRS acute and convalescent patients. IgG and IgM antibodies were detected by ELISA. Neutralizing antibodies were detected by neutralizing plaque reduction neutralization test. HFRSV RNA was extracted from serum of patients by Trizol method. TA test, determination of nucleotide sequence. Results The positive rate of serum IgM in HFRS patients was 82 2% (88 10 7), and the positive rate of IgG was 85 7% (6 6 77). Serum samples of 38 HFRS patients from two cities in the region were infected with house-based virus (SEO) and the other 6 were not typed. Fifteen of the 16 HFRS patients in another city were serologically infected , 1 is not stereotypes. The homology between HFRS virus and SEO in this area is more than 90%. Conclusions The Hantaan virus that caused the epidemic of HFRS in Yantai is a mixed virus mainly based on SEO.
