目的建立象皮HPLC指纹图谱分析方法,为其质量控制以及开发应用提供实验依据。方法用6 mol/L盐酸溶液水解象皮中的氨基酸,以异硫氰酸苯酯(PITC)为柱前衍生剂,用Hypersil ODS-2 C18色谱柱(250 mm×4.6 mm,5μm),梯度洗脱,体积流量为1.0 m L/min,检测波长为254 nm,柱温为40℃。利用“中药色谱指纹图谱相似度评价系统2004A版”对其进行相似度评价。结果建立了象皮HPLC指纹图谱共用模式,确定18个共有峰,并根据对照品指认17个共有峰,10批象皮药材具有较高的相似度,可反映象皮所含的17种氨基酸成分。结论首次建立了象皮的指纹图谱分析方法。该分析方法简便、快速,可为象皮质量的全面控制提供科学依据。
Objective To establish an HPLC fingerprint analysis method for elephant skin, providing experimental basis for its quality control and development and application. The 10 batches of elder herbs had higher similarity and could reflect 17 kinds of amino acids . Conclusion For the first time, the method of fingerprint analysis was established. The method is simple, rapid and can provide a scientific basis for the overall control of the elephant skin quality.
