目的:建立高效液相色谱法同时测定桑菊感冒片中绿原酸、木犀草苷和3,5-O-二咖啡酰基奎宁酸含量,以提高原质量标准。方法:样品用50%甲醇溶液提取,采用Phenomenex C18色谱柱(5μm,4.6 mm×250mm)分离;流动相为0.1%磷酸-乙腈,梯度洗脱;流速为1.0 mL·min-1;检测波长为348nm。结果:绿原酸、木犀草苷、3,5-O-二咖啡酰基奎宁酸分别在2.8128~140.6400μg·mL-1、0.9580~47.9023μg·mL-1、0.9879~49.3944μg·mL-1范围具有良好的线性;加标回收率分别为99.1%、100.9%、100.1%;RSD分别为1.5%、1.8%、1.8%(n=9)。结论:该方法简单易行,准确性和重复性好,可用于桑菊感冒片的质量控制。
OBJECTIVE: To establish a HPLC method for the simultaneous determination of chlorogenic acid, luteolin and 3,5-O-dis Caffeoylquinic acid in Sanguang cold tablets to improve the original quality standard. METHODS: The sample was extracted with 50% methanol solution and separated on a Phenomenex C18 column (5μm, 4.6 mm × 250mm). The mobile phase consisted of 0.1% phosphoric acid-acetonitrile and eluted with a gradient of 1.0 mL · min- 348nm. Results: The concentrations of chlorogenic acid, luteolin and 3,5-O-dicaffeoylquinic acid were in the range of 2.8128-140.6400μg · mL-1, 0.9580-47.9023μg · mL-1, 0.9879-49.3944μg · mL-1 The range was good linearity. The spiked recoveries were 99.1%, 100.9% and 100.1%, respectively. The RSDs were 1.5%, 1.8% and 1.8% respectively (n = 9). Conclusion: The method is simple, accurate and reproducible. It can be used for quality control of Sanguille cold tablets.
