目的了解甘肃省2009年风疹病毒的分子生物学特征,确定其基因型别。方法用Vero/Slam细胞从风疹暴发和散发患者的标本中分离风疹病毒,通过逆转录-聚合酶链反应(RT-PCR)和序列测定与分析鉴定其基因型。结果甘肃省首次分离出的3株风疹病毒鉴定为1E基因型。与2株世界卫生组织1E基因型的参考株(T14-CH-02株、M1-MAL-01株)形成了一个分支,均属于1E基因型,核苷酸同源性为99%~100%。结论此次疫情是由1E基因型风疹病毒引起的暴发疫情,此项研究为以后甘肃省风疹病毒的分子流行病学研究奠定了基础。
Objective To understand the molecular biological characteristics of rubella virus in Gansu Province in 2009 and determine its genotypes. Methods Vero / Slam cells were used to isolate rubella virus from specimens of patients with outbreaks and exudes of rubella and their genotypes were identified by reverse transcription-polymerase chain reaction (RT-PCR) and sequencing and analysis. Results The three rubella viruses isolated in Gansu Province for the first time were identified as 1E genotypes. It forms a branch with two WHO 1E genotype reference strains (T14-CH-02 and M1-MAL-01), belonging to the 1E genotype with nucleotide homology of 99% -100% . Conclusion The outbreak was caused by the 1E genotype rubella virus. This study laid the foundation for the molecular epidemiological study of rubella virus in Gansu Province in the future.
