目的观察幽门螺杆菌(Hp)感染后大鼠体内Th17细胞频数和IL-17水平的变化。方法使用Hp细菌悬液对30只大鼠灌胃,感染后3周、6周和9周分别处死大鼠,取大鼠的胃组织进行HE染色,观察胃黏膜病理改变情况,ELISA检测胃组织中IL-17含量。应用流式细胞术和反转录PCR技术检测大鼠脾脏淋巴细胞中Th17细胞频数和IL-17 mRNA水平。结果 Hp感染后,大鼠胃黏膜组织出现显著的病理改变;Hp感染时间越长,胃组织中IL-17含量越高;与正常对照组相比,Hp感染3周、6周和9周的大鼠脾脏淋巴细胞中Th17细胞频数显著增加,IL-17 mRNA表达水平亦显著升高,且各组间比较差异具有统计学意义。结论大鼠Hp感染不仅可导致胃黏膜损伤,胃组织IL-17含量增加,而且能够上调脾脏淋巴细胞中Th17细胞的频数和IL-17 mRNA水平。
Objective To observe the changes of Th17 cell frequency and IL-17 in rats after Helicobacter pylori (Hp) infection. Methods Thirty rats were inoculated intragastrically with Hp bacterial suspension and rats were sacrificed at 3, 6 and 9 weeks after infection respectively. The gastric tissues of rats were sacrificed and the pathological changes of gastric mucosa were observed. The gastric tissues were detected by ELISA In the IL-17 content. The frequency of Th17 cells and the level of IL-17 mRNA in splenic lymphocytes of rats were detected by flow cytometry and reverse transcription PCR. Results After Hp infection, gastric mucosal tissues of rats showed significant pathological changes. The longer the Hp infection time was, the higher the content of IL-17 in gastric tissues was. Compared with the normal control group, Hp infection was significant at 3, 6 and 9 weeks The frequency of Th17 cells and the expression of IL-17 mRNA in splenic lymphocytes of rats were significantly increased, and the difference between the groups was statistically significant. Conclusion Hp infection can not only cause gastric mucosal injury but also increase the content of IL-17 in the stomach tissue, and increase the frequency of Th17 cells and the level of IL-17 mRNA in splenic lymphocytes.
