目的:联合检测人涎腺腺样囊性癌组织中MMP-9、CD147的表达并分析其相关性,探讨其与临床病理参数间的关系,为腺样囊性癌的临床治疗和预后判断提供理论依据。方法:选择山西医科大学第一医院病理科存档的腺样囊性癌组织标本21例(癌组),正常涎腺组织6例(对照组)。21例腺样囊性癌分别依据临床分期、有无侵犯神经进行分组。运用免疫组织化学PV-9000二步法检测MMP-9、CD147,结果用SPSS 13.0软件分析。结果:MMP-9及CD147在腺样囊性癌组中的阳性表达率(分别为76.2%和81.0%)明显高于在对照组中的阳性表达率(分别为16.7%和16.7%),差异有统计学意义(分别为P<0.05和P<0.01)。Ⅲ+Ⅳ期腺样囊性癌病例MMP-9及CD147的阳性表达率(均为100.0%),明显高于Ⅰ+Ⅱ期病例(分别为44.4%和55.6%),差异有统计学意义(分别为P<0.01和P<0.05)。有无侵犯神经的病例组间比较,MMP-9及CD147的表达差异没有统计学意义(P>0.05)。MMP-9和CD147在腺样囊性癌组织中均呈阴性、弱阳性、阳性、强阳性表达者分别为4、5、7和3例,表达一致率为90.5%,Kappa值为0.870。结论:MMP-9和CD147均可作为反映腺样囊性癌细胞生物学行为的客观参考指标,其表达与临床分期密切相关,且二者之间的表达有正相关。
OBJECTIVE: To detect the expression of MMP-9 and CD147 in human salivary adenoid cystic carcinoma and to analyze the relationship between MMP-9 and CD147. To explore the relationship between MMP-9 and CD147 in clinical treatment and prognosis of adenoid cystic carcinoma Theoretical basis. Methods: Twenty-one cases of adenoid cystic carcinoma (NSCLC) and six cases of normal salivary gland (control group) were selected from Department of Pathology, the First Hospital of Shanxi Medical University. 21 cases of adenoid cystic carcinoma were divided according to clinical stage, with or without invasion of nerves. The immunohistochemical PV-9000 two-step method was used to detect MMP-9 and CD147, and the results were analyzed by SPSS 13.0 software. Results: The positive rates of MMP-9 and CD147 in adenoid cystic carcinoma (76.2% and 81.0%, respectively) were significantly higher than those in the control group (16.7% and 16.7%, respectively) There was statistical significance (P <0.05 and P <0.01, respectively). The positive rates of MMP-9 and CD147 in stage Ⅲ + Ⅳ adenoid cystic carcinoma were 100.0%, which were significantly higher than those in stage Ⅰ + Ⅱ (44.4% and 55.6%, respectively) P <0.01 and P <0.05, respectively). There was no significant difference in the expression of MMP-9 and CD147 between the two groups (P> 0.05). MMP-9 and CD147 in adenoid cystic carcinoma were negative, weak positive, positive, strong positive expression were 4, 5, 7 and 3 cases, the expression was 90.5%, Kappa value was 0.870. Conclusion: Both MMP-9 and CD147 can be used as an objective reference index to reflect the biological behavior of adenoid cystic carcinoma cells. The expression of MMP-9 and CD147 is closely related to the clinical stage, and there is a positive correlation between MMP-9 and CD147 expression.
