目的检测Beclin1、PTEN在鼻咽癌组织中的表达水平,分析自噬蛋白Beclin1与凋亡相关蛋白PTEN在鼻咽癌发生发展中的作用及临床意义。方法应用免疫组织化学方法检测50例鼻咽癌、20例癌旁黏膜和15例鼻咽黏膜慢性炎组织中Beclin1和PTEN蛋白表达,并分析其与临床分期、病理组织类型和转移的关系。结果Beclin1在鼻咽癌、癌旁黏膜和鼻咽黏膜慢性炎组织中阳性表达率分别为38.0%、65.0%、93.3%,PTEN的阳性表达率分别为34.0%、85.0%、93.3%,Beclin1和PTEN蛋白表达程度都与鼻咽癌临床分期、组织学分型和颈部淋巴结转移相关(P<0.05),且Beclin1与PTEN在鼻咽癌中的表达呈正相关(rs=0.946,P<0.05)。结论鼻咽癌组织中Beclin1、PTEN低表达与鼻咽癌发生、发展相关,检测其表达对研究鼻咽癌发生、发展有重要的作用,并可作为患者预后判断的重要标志物。
Objective To detect the expression of Beclin1 and PTEN in nasopharyngeal carcinoma and to analyze the role of autophagy Beclin1 and PTEN in the development of nasopharyngeal carcinoma and its clinical significance. Methods Immunohistochemistry was used to detect the expression of Beclin1 and PTEN protein in 50 cases of nasopharyngeal carcinoma, 20 cases of adjacent mucosa and 15 cases of chronic nasopharyngeal mucosal inflammation. The relationship between the expression of Beclin1 and PTEN protein was analyzed with clinical stage, pathological type and metastasis. Results The positive rates of Beclin1 expression in nasopharyngeal carcinoma, para-carcinoma and nasopharyngeal mucosa were 38.0%, 65.0% and 93.3%, respectively. The positive expression rates of Beclin1 were 34.0%, 85.0% and 93.3%, respectively. Beclin1 and The expression of PTEN protein was correlated with the clinical stage, histological type and lymph node metastasis of NPC (P <0.05). The expression of Beclin1 and PTEN was positively correlated with the expression of PTEN in nasopharyngeal carcinoma (rs = 0.946, P <0.05). Conclusion The low expression of Beclin1 and PTEN in nasopharyngeal carcinoma is correlated with the occurrence and development of nasopharyngeal carcinoma. Detecting the expression of Beclin1 and PTEN plays an important role in the study of the occurrence and development of nasopharyngeal carcinoma, and can be used as an important marker of prognosis.
