目的:建立HPLC同时分离测定了核桃仁中胞苷、尿苷、鸟苷、腺苷的含量。方法:采用Agilent TC-C18色谱柱(4.6 mm×250 mm,5μm),以甲醇-10 mmoL.L-1乙酸铵为流动相梯度洗脱,流速0.5 mL.min-1,检测波长260 nm,柱温30℃,测定了10个产地核桃仁中胞苷、尿苷、鸟苷、腺苷的含量。结果:4种核苷类成分在各自线性范围内,峰面积值与进样量呈良好的线性关系(r=0.999 3~0.999 9);回收率为98%~101%(RSD<3%),结论:方法简便快速、结果准确、重复性和稳定性好,可为核桃的含量测定提供参考。
OBJECTIVE: To establish simultaneous HPLC determination of cytidine, uridine, guanosine and adenosine in walnuts. METHODS: The mobile phase of methanol-10 mmoL.L-1 was eluted with Agilent TC-C18 column (4.6 mm × 250 mm, 5 μm) with a flow rate of 0.5 mL · min-1. The detection wavelength was 260 nm, The column temperature was 30 ℃, and the content of cytidine, uridine, guanosine and adenosine in 10 origin walnuts were measured. Results: The linear range of the four nucleoside components was linear with the injection volume (r = 0.999 3 ~ 0.999 9). The recoveries ranged from 98% ~ 101% (RSD <3%). Conclusion: The method is simple, rapid, accurate, repeatable and stable. It can provide a reference for determination of walnut content.
