目的 进一步了解与骨关节炎相关的关节软骨及游离体软骨细胞表型改变。方法 通过组化和免疫组化方法 ,观察纤维性胶原Ⅰ、Ⅱ、Ⅲ型在人正常和骨关节炎关节软骨及游离体中的表达。结果 在 10例骨关节炎关节软骨标本中 ,有 9例可在关节软骨的中层和深层的软骨细胞中显示强烈的Ⅱ、Ⅲ型胶原蛋白的阳性表达信号 ,而Ⅰ型胶原表达蛋白只在中下层及深层观察到 ,表达强度较Ⅲ型弱 ,表达区域部分与Ⅲ型重叠。在 10例骨关节炎游离体标本中 ,均可在游离体内部软骨细胞及基质中显示强烈的Ⅲ型胶原蛋白阳性表达信号。相同标本中Ⅰ型胶原蛋白仅在游离体内部软骨细胞中有表达 ,表达的阳性信号相对较低 ,未见明显的Ⅱ型胶原表达。在正常关节软骨中 ,Ⅱ型胶原的表达的信号较弱 ,未见Ⅰ、Ⅲ型胶原蛋白表达信号。结论 在骨关节炎关节软骨及游离体中 ,一定区域内的软骨细胞表型发生了改变 ,表达合成了Ⅲ型胶原和Ⅰ型胶原 ,可能在骨关节炎发病机制中发挥重要作用。
Objective To further understand the changes of articular cartilage and free-form chondrocytes associated with osteoarthritis. Methods The expression of fibrillar collagen type Ⅰ, Ⅱ and Ⅲ in human articular and osteoarthritis articular cartilage and free body was observed by immunohistochemistry and immunohistochemistry. Results Of the 10 osteoarthritis articular cartilage specimens, 9 showed strong expression of type II and type III collagen in the middle and deep cartilage of articular cartilage, while type I collagen expressed only in the middle The lower and deeper layers were observed that the expression intensity was weaker than that of type III and the expression regions overlapped with type III. In 10 cases of osteoarthritis free specimens, both in vivo free chondrocytes and matrices showed strong expression of type Ⅲ collagen positive signals. Type I collagen in the same specimen was expressed only in free internal chondrocytes, the positive signal was relatively low, and no obvious expression of type II collagen was observed. In normal articular cartilage, the expression of type Ⅱ collagen signal is weak, no type Ⅰ, Ⅲ collagen expression signal. Conclusion In the articular cartilage and free body of osteoarthritis, the phenotype of chondrocytes in a certain area is changed, and the expression of type Ⅲ collagen and type Ⅰ collagen may play an important role in the pathogenesis of osteoarthritis.
