本文建立了一种以吸附在硝酸纤维素纸上的日本血吸虫循环抗原为靶子,用生物素化的抗虫源性和虫卵源性的高效特异的多克隆抗体为探针检测血吸虫循坏抗原的方法。该法利用生物素一亲和素的多级放大作用,并用聚丙烯酰胺凝胶颗粒浓缩血清及解离复合物等方法来提高敏感性。显示这种探针阳性率高、假阳性率低用于诊断具有较好的敏感性和特异性。该探针与DNA探针比较具有简便、快速、不需特殊仪器等优点,可望成为确诊病人和考核疗效的工具。
In this paper, we established a method to detect the circulating antigen of Schistosoma japonicum by using the antigen of Schistosoma japonicum adsorbed on the nitrocellulose paper as the probe, using biotinylated and egg-derived polyclonal antibodies as probes Methods. This method utilizes the multi-stage amplification of biotin-avidin and uses polyacrylamide gel particles to concentrate serum and dissociate complexes to increase sensitivity. This probe shows a high positive rate, low false positive rate for diagnosis with good sensitivity and specificity. Compared with the DNA probe, the probe is simple, rapid and requires no special instruments. It is expected to become a tool for diagnosing patients and evaluating the curative effect.
