目的本研究旨在探讨microRNA(miR)-187在胃癌组织中的表达情况,并分析miR-187表达的改变与胃癌分化程度和TNM分期之间的关系。方法 32例胃癌及相应癌旁正常胃黏膜组织标本提取RNA后,采用茎环逆转录实时荧光定量PCR(real time-PCR)方法,分别检测miR-187在胃癌及癌临近正常胃黏膜组织中的表达量,并进一步观察miR-187表达的变化同胃癌分化程度以及TNM分期之间存在的联系。结果胃癌及癌旁正常胃黏膜组织中均可以检测到miR-187,且与癌临近的正常胃黏膜组织相比,miR-187的表达量发生显著的改变,其在胃癌组织中表达出现显著性的下降(P<0.05)。胃癌组织中的miR-187表达量与胃癌的分化程度(P<0.05)及TNM分期(P<0.05)均显著相关。结论在胃癌组织及癌临近胃黏膜组织中,miR-187的表达呈明显减少的趋势,差异具有统计学意义;且胃癌组织中miR-187表达的减少与胃癌的分化程度以及TNM分期均呈显著的相关性。以上研究结果表明,miR-187表达的减少与胃癌的发生、发展密切相关。
Objective This study aimed to investigate the expression of microRNA (miR) -187 in gastric cancer and to analyze the relationship between the changes of miR-187 expression and the differentiation of gastric cancer and TNM staging. Methods RNA was extracted from 32 specimens of normal gastric mucosa and adjacent gastric mucosa. Real-time PCR was used to detect the expression of miR-187 in gastric cancer and adjacent normal gastric mucosa And to further observe the relationship between the change of miR-187 expression and the degree of gastric cancer differentiation and TNM staging. Results miR-187 could be detected in gastric cancer and adjacent normal gastric mucosa tissues, and the expression of miR-187 was significantly changed compared with the normal gastric mucosa adjacent to cancer, which was significant in gastric cancer (P <0.05). The expression of miR-187 in gastric cancer tissues was significantly correlated with the differentiation of gastric cancer (P <0.05) and TNM stage (P <0.05). Conclusions The expression of miR-187 in gastric cancer tissues and adjacent cancerous tissues has a tendency to decrease, the difference is statistically significant. The decrease of miR-187 expression in gastric cancer tissues is associated with the differentiation of gastric cancer and TNM staging Relevance. The above results show that the reduction of miR-187 expression is closely related to the occurrence and development of gastric cancer.
